1.1 Narrative summary
Excretion of radioactivity was rapid with 80 % of the dose being recovered by 24 hours after dosing and a mean total recovery of 87.6 % to 48 hours. Approximately 61 % of the dose was recovered as AZD5672, 31 % in urine, 28 % in faeces and 2 % in bile.
The major circulating component, corresponding to 84.6 % of the total radioactivity, wasidentified as AZD5672 with the major metabolite being an oxidised metabolite, 4.6 % of the total radioactivity. Two further minor metabolites, both singly oxidised, were present but not quantifiable.
Approximately 30 % of the radioactive dose was recovered in the faeces, predominantly as AZD5672, suggesting direct secretion into the intestine.
Several metabolites were identified in bile with the major component, 3.2 % of the dose, being an oxidised glucuronide, with the oxidation present on the difluorophenyl ring. Other components present were AZD5672 (1.6 %), and two oxidised metabolites (2.4 and 1.0 % of dose). Minor metabolites identified included singly and doubly oxidised products, an Ndealkylated
amine and a defluorinated, oxidised glycyl-cysteinyl conjugate, each
corresponding to less than 1 % of the dose. A number of other oxidised, conjugated and cleaved metabolites were present but were not quantifiable.
The largest component identified in urine samples was AZD5672, corresponding to 31.0 % of the dose. No urinary metabolite constituted more than 1 % of the dose and their structures were similar to those present in bile, including singly and doubly oxidised products, an N-dealkylated amine and a defluorinated, oxidised glycyl-cysteinyl conjugate.
